Indications/Uses
In conditions where inhibition of gastric acid secretion may be beneficial, including aspiration syndromes, dyspepsia, GERD, PUD, & Zollinger-Ellison syndrome.
Dosage/Direction for Use
IV Adult 40 mg over a period of 20-30 min by IV infusion, may also be given by slow IV inj. Childn 1 mth-12 yr 500 mcg/kg (to a max of 20 mg) once daily; may be increased to 2 mg/kg (to a max of 40 mg) once daily if needed. Hepatic impairment 10-20 mg daily.
Special Precautions
Risk of GI infections. May mask symptoms & delay diagnosis of malignancy. Exclude possibility of malignancy before therapy to patients w/ gastric ulcers. Hepatic impairment. Pregnancy & lactation.
Adverse Reactions
Headache, diarrhoea, & skin rashes. Pruritus, dizziness, fatigue, constipation, nausea & vomiting, flatulence, abdominal pain, arthralgia & myalgia, urticaria, & dry mouth.
Drug Interactions
May affect exposure w/ inhibitors or inducers of CYP2C19 & CYP3A4. May alter metabolism of some drugs metabolized by CYP2C19 & CYP3A4. May prolong elimination of diazepam, phenytoin, & warfarin. Reduced absorption of dasatinib, ketoconazole, & itraconazole. Increased plasma conc of both voriconazole & omeprazole. Reduces exposure to atazanavir.
